Monitoring the Interaction of SARS-CoV-2 with ACE-2 and the Resulting Immune Response with Flow Cytometry


Angiotensin Converting Enzyme 2 (ACE-2) is a widely expressed receptor that is utilized by coronaviruses and used as an entry port into cells. SARS-CoV-2 binds to ACE-2 through its Spike 1 protein RBD, and this receptor binding is the first step in viral entry. Antibody or small molecule blockade of this interaction could be a viable therapeutic option; however, research is hampered by biosafety considerations. To progress development of COVID-19 therapeutics, we developed a flow cytometry-based in vitro assay for evaluating the effectiveness of antibodies and/or small molecules to block the binding of the Spike protein to ACE-2. Infection with SARS-CoV-2 results in a wide range of immune system responses. Flow analysis of expression of ACE-2 on monocytes or macrophage subsets may provide clues to help determine disease progression or pathogenesis.

Viewers will gain insights into ACE-2 detection and blocking of the ACE-2/SARS-CoV-2 RBD interaction in an in vitro flow cytometry-based assay. We will also be looking at flow cytometry analysis of ACE-2 expression on monocytes, and immune cell response to IL-6.

Dr. Jody Bonnevier is a Manager in the Antibody Development Department at R&D Systems in Minneapolis MN. Jody received her PhD in Microbiology, Immunology, and Cancer Biology at the University of Minnesota, and has over 10 years of experience developing novel antibodies and reagents, with a focus on flow cytometry and cell selection product development.
