Multiplexed Protein Detection in Heterogeneous Neural Cell Subpopulations with Single-Cell Westerns

Single-Cell Western blotting can measure protein expression in neurons and other neural cells that may be hard to examine using flow cytometry or to quantify using immunofluorescence. In this talk, we discuss how the Single-Cell Western blotting platform, Milo, can be used for multiplexed protein detection in heterogeneous neural cell subpopulations. Milo identifies subpopulations like neural progenitor cells, astrocytes, neurons, and oligodendrocytes in a mixed population and can quantify differentiation efficiency and protein expression heterogeneity in neural samples.

As cell and gene therapy options to combat neurodegenerative disease continue to show promise, new focus is being directed toward more efficient, innovative, and cost-effective methods for commercial-scale manufacturing. Using induced pluripotent stem cell (iPSC)-derived neurons as a case study, this session will discuss the importance of raw material optimization from research to translation, including early incorporation of GMP small molecules, growth factors and media supplements.

Learn how the RNAscope™ and BaseScope™ in situ hybridization (ISH) assays can allow you to spatially map gene expression targeting RNA sequences directly in fresh frozen, fixed frozen, and formalin-fixed paraffin-embedded (FFPE) cells and tissues with single-cell resolution. We will focus on the multiplexing capabilities of the RNAscope Multiplex Fluorescent assay and the new RNAscope HiPlex assay, with simultaneous detection of up to 12 targets, as well as the detection of specific splice variants using the BaseScope assay.
